Lodopin-V: A Comprehensive Solution for Hypertension Management

Lodopin-V is a combination tablet designed to help manage high blood pressure. It contains amlodipine, a dihydropyridine calcium channel blocker, valsartan, an angiotensin II receptor blocker, and hydrochlorothiazide, a thiazide diuretic. These three components work together to lower blood pressure by blocking the contractile effects of calcium on cardiac and vascular smooth muscle cells, inhibiting the vasoconstrictive and sodium-retaining effects of angiotensin II, and promoting the excretion of sodium and chloride in the kidneys.

How It Works

• Amlodipine: Blocks the contractile effects of calcium on cardiac and vascular smooth muscle cells, leading to relaxation of these muscles and a subsequent decrease in blood pressure.

Dosage and Administration

The recommended dose of Lodopin-V is one tablet per day. The majority of its effect is attained within two weeks. It may be administered with or without food and can be used as add-on therapy for patients not controlled on monotherapy. For initial therapy, start with a lower dose (5/160/12.5 mg) and titrate upwards as necessary to a maximum of 10/320/25 mg once daily.

Important Considerations

• Contraindications: Known hypersensitivity to any component.
• Pregnancy and Lactation: Discontinue use if pregnancy is detected, and breastfeeding is not recommended.
• Geriatric and Pediatric Use: Not recommended for initial therapy in geriatric patients or pediatric use.
• Renal Impairment: Safety and effectiveness in patients with severe renal impairment have not been established.

Precautions

• Potassium Levels: Use with caution when taking potassium supplements, potassium-sparing diuretics, or salt substitutes containing potassium.
• Renal Artery Stenosis: Use with caution in patients with unilateral or bilateral renal artery stenosis or stenosis to a solitary kidney.

Overdose

Limited data are available related to overdosage in humans. The most likely manifestations of overdosage would be hypotension and tachycardia; bradycardia could occur from parasympathetic (vagal) stimulation.
